چکیده

an ice core on the Antarctic Plateau as part of a long-term European ice core drilling collaboration. Michael Bock et al. found that seasonal floods and tropical wetlands likely controlled atmospheric methane concentrations during the current and two older interglacial periods as well as their preceding glacial maxima, and that geologic methane emissions may have a constant but minor role in atmospheric methane fluctuations. According to the authors, increased fire activity may have influenced methane stable isotope levels throughout the last 25,000 years. See the article by Michael Bock et al. on pages E5778–E5786. Antimalarials inhibit hematin crystallization by unique drug-surface site interactions.

In malaria pathophysiology, divergent hypotheses on the inhibition of hematin crystallization posit that drugs act either by the sequestration of soluble hematin or their interaction with crystal surfaces. We use physiologically relevant, time-resolved in situ surface observations and show that quinoline antimalarials inhibit β-hematin crystal surfaces by three distinct modes of action: step pi...

Zinc protoporphyrin IX binds heme crystals to inhibit the process of crystallization in Plasmodium falciparum.

The intraerythrocytic Plasmodium falciparum parasite converts most of host hemoglobin heme into a nontoxic heme crystal. Erythrocyte zinc protoporphyrin IX, normally present at 0.5 microM, which is a ratio of 1:40,000 hemes, can elevate 10-fold in some of the anemias associated with malaria disease protection. This work examines a binding mechanism for zinc protoporphyrin IX inhibition of heme ...

Mechanisms of hematin crystallization and inhibition by the antimalarial drug chloroquine.

Hematin crystallization is the primary mechanism of heme detoxification in malaria parasites and the target of the quinoline class of antimalarials. Despite numerous studies of malaria pathophysiology, fundamental questions regarding hematin growth and inhibition remain. Among them are the identity of the crystallization medium in vivo, aqueous or organic; the mechanism of crystallization, clas...

The malaria digestive vacuole.

During the development of malaria parasites within human erythrocytes, the fusion of digestive vesicles gives rise to a large digestive vacuole (DV). This organelle, which is maintained at low pH, processes 60-80 percent of the erythrocyte hemoglobin to provide a pool of amino acids that is crucial for parasite growth and development. During proteolysis, heme is released from hemoglobin as a t...

Heme Polymerization Inhibitory Activity (HPIA) of N-alkyl and N-benzyl-1,10 Phenanthroline Derivatives as Antimalaria

Malaria parasite metabolizes hemoglobin and detoxifies the resulting toxic ferriprotoporphyrin IX by polymerization into a crystal insoluble pigment, known as hemozoin. A polymer identical to hemozoin, β-hematin, can be obtained in vitro from hematin at acidic pH. Quinoline-containing antimalarials (e.g. chloroquine) inhibit the formation of polymer. Thus, heme polymerization is an essensial an...
